چکیده مقاله:

BMP15 and GDF9 are two oogenesis specific genes play a pivotal role in female fertility in mammals and potential for improvement of prolificacy in marker-assisted selection. The aim of present research was to investigate the variation and association between BMP15 and GDF9 polymorphism and litter size in Markhoz goats. The sequence variability of the different amplified fragments utilized for genotyping was further investigated by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP), polymerase chain reaction-single strand conformation polymorphism (PCR-SSCP) and sequencing. Sequencing technology allowed the detection of two nucleotide variations: A both non-functional mutation (G/A) transition at nt 1026 known as P3) and a synonymous mutation at 1049 position which for the first time was found at the BMP15 gene. Also, GDF9 gene exhibited a polymorphism on BcnI site (1159 bp) within exon 2 of amplified PCR products (glutamine/proline). Our analysis using general linear model (GLM) statistical methods showed litter size was significantly influenced by genotypes of GDF9 (P

Single Nucleotide Polymorphisms (SNPs) of GDF9 Gene in Bahmaei and Lak Ghashghaei Sheep Breeds and Its Association with Litter Size

Growth differentiation factor 9 (GDF9) belong to the superfamily of transforming growth factor β that is highly expressed in growing ovarian follicles of oocyte, and it has been strongly related to fecundity traits in sheep. Therefore, the GDF9 gene could serve as a genetic marker for improvement of reproductive performance in sheep. Therefore, the aim of this study was to invest...

Novel single nucleotide polymorphisms of GnRHR gene and their association with litter size in goats

In the present study, the polymorphisms of gonadotropin-releasing hormone receptor (GnRHR) gene were analysed as a genetic marker candidate for litter size in 720 individuals from Shaanan goats (SG) and Boer goats (BG). Two alleles (A and C), two observed genotypes (AA and AC), and single nucleotide polymorphisms (SNPs) were detected. The frequencies of alleles A and C in two goat breeds were 0...

Polymorphisms in GDF9 Gene and Its Relationship with Litter Size in Five Breeds of Black Goats

In this study, we investigated the relationship between the genetic polymorphism of growth differentiation factor 9 (GDF9) genes and the litter size in 384 individuals of five breeds of black goats. Four pairs of primers were designed to detect single nucleotide polymorphism of GDF9 gene in goats by PCR-SSCP. The least square was used to analyze the relation between different genotypes and the ...

The association of two single nucleotide polymorphisms (SNPs) in growth hormone (GH) gene with litter size and superovulation response in goat-breeds

Two active mutations (A 781 G and A 1575 G) in growth hormone (GH) gene, and their associations with litter size (LS), were investigated in both a high prolificacy (Matou, n = 182) and a low prolificacy breed (Boer, n = 352) by using the PCR-RFLP method. Superovulation experiments were designed in 57 dams, in order to evaluate the effect of different genotypes of the GH gene on superovulation r...

Association of Candidate Single Nucleotide Polymorphisms Related to Candidate Genes in Patients With Schizophrenia

Introduction: Schizophrenia is a chronic heterogenic neurodevelopment disorder. Many genes interfere in the development of SCZ. All four genes, NrCAM, PRODH, ANK3, and ANKK1, which were evaluated in this study, were previously reported to be associated with Schizophrenia. The NrCAM contributes to creating cognitive deficiencies through the CAM’s signaling pathway. PRODH plays a vital role in cr...

Novel Single Nucleotide Polymorphisms (SNPs) in Intron 2 and Exon 3 Regions of Leptin Gene in Sumba Ongole Cattle

The bovine leptin (LEP) gene was widely used as a candidate gene for molecular selection to improve productivity traits of cattle. This study was carried out to identify single nucleotide polymorphisms (SNPs) in the LEP gene of Sumba Ongole (SO, Bos indicus) cows using sequencing method. A total of 31 animals were used in this study for analyses. Research showed that total of 16 SNPs w...
